Efikasi Filtrat Kasar Saponin Biji Keben (Barringtonia asiatica) Terhadap Bakteri Vibrio harveyi

Abstract

Pengendalian Vibrio harveyi, patogen penyebab luminous vibriosis, masih bergantung pada antibiotik konvensional, yang memicu resistensi antimikroba (AMR). Saponin dari biji Barringtonia asiatica (keben) telah dilaporkan memiliki aktivitas antibakteri. Penelitian ini bertujuan mengevaluasi efektivitas filtrat kasar saponin B. asiatica pada berbagai dosis terhadap V. harveyi. Pengujian dilakukan menggunakan Rancangan Acak Lengkap (RAL) dengan enam perlakuan (A=0,7 ppm, B=0,9 ppm, C=1,1 ppm, D=1,3 ppm, E=1,5 ppm, dan Kontrol=0 ppm), masing-masing tiga ulangan. Kultur V. harveyi (1x106 sel/mL) diinkubasi selama 24 jam bersama filtrat. Efektivitas diukur melalui Total Bacterial Count (TBC) pada media TCBSA dan dihitung sebagai Persentase Penghambatan (%) serta Log Reduction. Filtrat kasar saponin secara signifikan memengaruhi pertumbuhan V. harveyi (p < 0,05). Efektivitas tertinggi dicapai pada dosis E (1,5 ppm), menghasilkan 90,00% penghambatan dan 0,98 Log Reduction. Filtrat kasar saponin biji B. asiatica merupakan agen antibakteri yang efektif terhadap V. harveyi pada dosis tinggi. Filtrat ini memiliki potensi besar sebagai alternatif biokontrol berkelanjutan dalam akuakultur.
